There are a couple things that you need to do to get Exchange 2007 to work, I imagine that you would have to do something similar for 2010. For example the POP3 Service isn't configured to start by default.
Goto Services and make sure that POP3 is running. If it isn't you can start it and set it to run automatically.
Also on Exchange 2007 we had to actually configure POP3 and IMAP to not require SSL. (2007 was when they started to require an SSL connection for POP3 and IMAP and thus you either have to use SSL and the appropriate SSL port numbers or configure exchange to not require SSL.)
Also make sure that the user is enabled for POP3 or IMAP in their user profile. I ended up giving up on making POP3 work and use IMAP.
